The biosynthesis of oligosaccharide-lipids. human milk oligosaccharides of an An oligosaccharide-P-P-lipid has been isolated from porcine liver by extraction with organic solvents and purified by chromatography on silica gel and DEAE-cellulose. The purified oligosaccharide-lipid was shown to contain mannose and N-acetylglucosamine in an approximate ratio of 11 and our results suggest that the major oligosaccharide component in the preparation was a tetrasaccharide with the composition (Man)2 (GlcNAc)2. When the oligosaccharide-lipid was incubated with GDP-[14C]mannose and a solubilized enzyme preparation from rabbit liver in the presence of MgCl2, three radioactive products could be isolated. Order immediately in the products could be identified as a penta-, a hexa-, and a heptasaccharide. These products were formed by the stepwise addition of mannose to the growing oligosaccharide chain and GDP-mannose was indicated as the glycosyl donor in each reaction.

The milk protein α-casein functions as a tumor suppressor via activation of STAT1 signaling, effectively preventing breast cancer tumor growth and Bonuccelli G(1), Castello-Cros R, Capozza F, Martinez-Outschoorn UE, Lin Z, of Stem Cell Biology & Regenerative Medicine, Thomas Jefferson University, Here, we identified the milk protein α-casein as a novel suppressor of tumor growth and metastasis. Briefly, Met-1 mammary tumor cells expressing α-casein showed a ~5-fold reduction in tumor growth and a near -fold decrease in experimental metastasis. To identify the molecular mechanism(s), we performed genome-wide transcriptional profiling. Interestingly, our results show that α-casein upregulates gene transcripts associated with interferonSTAT1 signaling and downregulates genes associated with stemness. These findings were validated by immunoblot and FACS analysis, which showed the upregulation and hyperactivation of STAT1 and a decrease in the number of CD44(+) cancer stem cells. These gene signatures were also able to predict clinical outcome in human breast cancer patients. Thus, we conclude that a lactation-based therapeutic strategy using recombinant α-casein would provide a more natural and non-toxic approach to the development of novel anticancer therapies.

The chemistry of oligosaccharide ligands of selectins significance for the development of new immunomodulatory medicines.Glycomic work-flow for analysis of mucin O-linked oligosaccharides. The high-throughput analysis of the glycosylation of high molecular weight proteins, such as mucins, has been the aim of glycomics initiatives for the last decade. Here, we present a work-flow for the efficient and reproducible analysis of reduced oligosaccharides from a typical mucin sample. This work-flow can be applied to any similar samples of oligosaccharides. We include recently developed bioinformatic procedures for the statistical analysis of sample sets. These procedures can be applied in any laboratory environment, using free programs that are platform independent.

The scripts are explained and can be adjusted to suit the individual experiment. Finally, a number of example results are given to highlight the use of the statistical analysis in a biological Synthesis of heparan sulfate tetrasaccharide as a substrate for human Takeda N(1), Ikeda-Matsumi R, Ebara-Nagahara K, Otaki-Nanjo M, Taniguchi-Morita Regiospecifically sulfated heparan sulfate tetrasaccharide, GlcAβ-GlcN(NS6S)α-GlcAβ-GlcN(NS6S)α was first synthesized as an octyl glycoside. Chemical, enzymatic and chromatographic analyses show that each vitellin contains variably processed high mannose type oligosaccharides. While all have a common size range they occur as two distinct classes based on the proportion of individual saccharides present. Oligosaccharide size distribution is not a characteristic of an individual animal but of the species. Because oligosaccharide heterogeneity also occurs in B.
